RESEARCH EXPERIENCE:
During my research career, I have worked on following projects:

  • Implication of Akt signaling in HIV-1 persistence and formation of viral reservoirs in CD4+ T cells during chronic viral infection
  • HIV-1 pathogenesis in HIV-1/HCV coinfection .
  • Oncomodulatory role of HCMV in liver cancer and role of HDAC inhibitors in rheumatoid arthritis:

Molecular Biology:
General: DNA/RNA isolation, PCR & Real time PCR, Recombinant DNA technology/Gateway cloning, Transgenesis in
Drosophila melanogaster, Mapping of transgene.
Protein: Protein expression and protein purification, protein-protein interaction (GST-pull down assay,
immunoprecipitation), DNA- protein interaction (electrophoretic mobility shift assay), isolation of protein from cytoplasmic
fraction, mitochondrial fraction and nuclear fraction, Immunoblotting/SDS-PAGE.
 

Virology:
FMD viral isolation from clinical samples, genotyping of FMDV, HIV-1 drug testing, HIV-1/HCV viral propagation,
quantification and infection using molecular clones; viral RNA isolation, quantification of HIV-1 reservoirs, Roche
molecular Diagnostic (COBAS-AmpliPrep and Taqman 48 HIV Test)
 

Cell Biology:
Animal cell culture: Primary cells- isolation of PBLs and Monocytes from buffy coats, culturing human monocytederived macrophages (MDM), and monocytic cell lines (U937), hepatocytes etc; protein expression and purification in
mammalian cell lines, siRNA transfection,
Flow cytometry: Apoptosis, Immunophenotyping, Fluorescent protein analysis, Cell sorting
Immunological assays: ELISA, Cell proliferation/toxicity assays, Fluorescent microscopy, immunohistochemical
staining.

Bioinformatics Skills
Comparative genomics, Homology modeling, Phylogentic tree construction, Multiple sequence alignment, BLAST,
protein/DNA analysis tools, Primer designing, Familiar with “Deep view”, “Bioedit” and “Jalview”.
Computer skills: Competent with most of Microsoft office programs and knowledge of the principal software for
molecular biology.
